3D structure thermostable cellobiohydrolase

Abstract

X-ray crystallography was used to determine the 3D structure of the cellobiohydrolase IB (CBH IB) enzyme from Talaromyces emersonii PDB 1Q9H. The ce17 gene encoding CBH IB was isolated and used to transform Escherichia coli to produce the enzyme. The recombinant enzyme was almost inactive against methylumberiferyl-cellobioside and chloronitrophenyl-lactoside. Refolding of the enzyme after denaturing it with urea restored partial activity.
